abstract | Production of medium chain length poly-3-hydroxyalkanoatesn(mcl-PHAs) in Escherichia coli is reported for the firstntime. To produce PHA in E . coli , a PHA polymerase encodingngene ( phaC2 ) of Pseudomonas oleovorans GPo1 was introducedninto various E . coli strains and fatty acid oxidation mutantsnthereof. Of these, E . coli fadA/B , E . coli fadRfadA or E.n coli fadRfadB mutants, which are defective in specific stepsnof the fatty acid β-oxidation pathway, allowed production ofnmcl-PHAs from fatty acids ranging from octanoate to octadecanoate.nMaximum amounts of about 10% PHA of total cell drynweight (w/w) were found. The predominant monomers found innthese PHAs were always the same, independently of the chainnlength of the supplied fatty acids: 3-hydroxyoctanoate forneven chain length fatty acids and a combination of 3-hydroxynonanoatenand 3-hydroxyheptanoate for odd chain length fattynacids. Since the monomer composition of the produced polymernwas independent of the fatty acid chain length, β-oxidationnoccurs in these β-oxidation mutants. The polymer may benhydrolyzed to yield interesting optically active monomers andntheir esters. |
