abstract There is described a new process for manufacturing the nucleosidic derivative 1-β-D-arabinofuranosylcitosyne (cytarabine, Ara-C), according to which 1-β-D-arabinofuranosyluracyl (Ara-U) is treated, under reduced pressure and under heating, with hexamethyldisilazane and a C 1 -C 3 acylamide. n After evaporating the solvent and treatment with methyl alcohol and aqueous ammonia, the reaction mixture is subjected to purification on an acidic resin, thereby obtaining the relevant product in high yields.
