titleOfInvention | METHOD OF OBTAINING POLYPEPTIDES |
abstract | The present invention relates to methods for optimizing protein coding sequences for expression in predetermined host cells. In these methods, genetic algorithms are used to optimize sequences encoding a given amino acid sequence for the suitability of individual codons and / or for the suitability of codon pairs. When creating new variants of sequences and subsequent selection of the best variants, the algorithm iterates until variants of the coding sequences reach the minimum value of the suitability of individual codons and / or suitability of codon pairs. The invention also relates to a computer including a processor and a storage device (memory), the computer being configured to read from and write to the memory, the memory including data and instructions designed to give the processor the ability to execute genetic algorithms to optimize the suitability of individual codons and / or suitability of codon pairs. The invention also relates to nucleic acids containing a coding sequence for a given amino acid sequence, the coding sequence being optimized for the suitability of individual codons and / or the suitability of codon pairs for a given host by the methods of the invention, as well as host cells containing such nucleic acids, and methods for producing polypeptides and other fermentation products in which these host cells are used. |
