abstract |
The present disclosure provides a method for reducing or eliminating nucleic acid contamination of amplification products during second-generation sequencing and application in the detection of novel coronavirus 2019-nCoV. Aiming at the common false positive problem of next-generation sequencing platforms, the library is constructed through the long-range amplification-amplicon fragmentation scheme, and dUTP and nucleases that can specifically recognize dUTP are used in the long-range amplification system. The method of the present disclosure can reduce or eliminate false positives caused by contamination of amplification products, and improve the accuracy of detection. |
