abstract The invention discloses a culture medium and a culture method for primary culture of tumor-infiltrating mast cells. The culture medium comprises the following components: 0.05-1v% PVA, 0.5-2v% ITSX, 80-120U/mL penicillin, 0.05‑1mg/mL streptomycin, 50‑60μM/L 2‑mercaptoethanol, 80‑120ng/mL SCF, 80‑120ng/mL IL‑6, and the balance is IMDM medium. The culture method includes the following steps: S1. Washing fresh tumor tissue, and then cutting it into small pieces of 2-4 mm 3 ; S2. Making the tissue piece obtained in S1 into a single cell suspension, then adding CD117 + immunomagnetic beads to incubate, washing the cells, Centrifuge and resuspend, and select mast cells by immunomagnetic bead sorting system; S3. Resuspend the mast cells obtained from S2 in the medium used for primary culture of tumor-infiltrating mast cells, and then transfer them to a well plate, and put them in a well plate. Culture in a 37°C, 5% CO 2 incubator, and perform half-volume medium changes 2-3 times a week. The efficiency of amplifying the mast cells by the method of the present invention is significantly improved, and the cost is significantly reduced at the same time.
