abstract | The invention provides a rice bidirectional single-base edited co-transcription unit gene ABE-CBE system, a vector and application thereof, wherein the co-transcription unit gene ABE-CBE comprises an adenine deaminase coding gene TadA and a cytosine deaminase coding gene CDA, and coding sequences of a co-transcription unit gene mutant TadA and LjCDAL1 are constructed in the same expression vector. The invention fuses the expression vector containing the cytosine base editor and the adenine base editor to cause the nucleotide replacement of specific loci of a genome, and experiments prove that the fusion editing tool designed by the invention is used for constructing the plant expression vector and further constructing the rice targeted editing vector, the mutual replacement efficiency of A: T to G: C on DNA of the specific loci of the rice is improved after the vector is introduced into rice cells, the simultaneous replacement of C to T and A to G can be realized on the specific loci of the genes, the application range is expanded, and the generation of off-target is reduced. The fused A-C base editing system has wider application prospect. |
