The present disclosure relates to bifunctional compounds that are useful as modulators of Kirsten rat sarcoma proteins (target proteins). In particular, the present disclosure relates to bifunctional compounds containing at one end a Von Hippel-Lindau, cereblon, inhibitor of apoptotic protein, or a mouse bibody 2 homolog ligand bound to the respective E3 ubiquitin ligase, and another One end contains a moiety that binds to the target protein, so that the target protein is placed in the vicinity of the ubiquitin ligase to effect degradation (and inhibition) of the target protein. The present disclosure exhibits broad pharmacological activities related to degradation/inhibition of target proteins. Diseases or disorders caused by aggregation, accumulation and/or overactivation of target proteins are treated or prevented with the compounds and compositions of the present invention. |
