abstract | The invention discloses a linear chain malto-oligosaccharide generating enzyme mutant with improved malto-hexaose production capacity, belonging to the technical field of enzyme engineering. The straight chain malto-oligosaccharide generating enzyme mutant takes 5% (w/w) corn starch solution as a substrate, the percentage content of the malto-oligosaccharide in the product is respectively 36.10% (G109N), 42.40%, (G109D) and 38.99% (G109F), the maltose-oligosaccharide generating enzyme mutant is 1.10, 1.29 and 1.18 times of that of a wild type (32.93%), and the substrate conversion rate is improved to more than 79%. Even under higher substrate concentration (20% of corn starch, w/w), the mutant G109D can still effectively hydrolyze starch to produce high-concentration maltohexaose, the percentage content of the maltohexaose in the product is 44.69%, the high product purity and the substrate conversion rate can effectively reduce the production and processing cost of the high-purity straight-chain maltooligose syrup, and the high-purity straight-chain maltooligose syrup has industrial application value. |
