The present invention relates to methods for detecting DNA modifications and protein binding by single-molecule manipulation. In particular, the invention relates to methods for determining whether a protein binds to a specific DNA sequence. This method is particularly useful for identifying DNA sequence modifications (eg, methylation) through the binding of proteins (eg, antibodies) that specifically recognize these modifications, and also for identifying the binding sequences of various proteins on DNA. |
