The present description relates to imide-based compounds, including bifunctional compounds comprising them, which are useful as modulators of targeted ubiquitination, in particular inhibitors of a variety of polypeptides and other proteins which are degraded and/or otherwise inhibited by bifunctional compounds according to the present invention. In particular, the present specification provides compounds containing a ligand on one end that binds to cereblon E3 ubiquitin ligase and a moiety on the other end that binds a target protein such that the target protein is located proximal to the ubiquitin ligase to effect degradation (and inhibition) of the protein. Compounds can be synthesized that exhibit a wide range of pharmacological activities consistent with degradation/inhibition of virtually any type of targeted polypeptide. |
