abstract | The invention discloses an alkaline xanthine dehydrogenase and its application in a detection kit. The protein disclosed in the invention includes a large xanthine dehydrogenase subunit and a small xanthine dehydrogenase subunit. The alkaline xanthine dehydrogenase provided by the invention is the first reported xanthine dehydrogenase of the genus Acinetobacter baumannii confirmed by experiments. The alkaline xanthine dehydrogenase provided by the invention can be adapted to samples under alkaline determination conditions, has higher substrate affinity and the highest catalytic efficiency than existing reports, and can more sensitively degrade and detect hypoxanthine/xanthine And PNP enzyme and 5'-nucleotidase, combined with PNP enzyme to detect inorganic phosphate and adenosine deaminase, is beneficial to improve enzyme catalytic efficiency, reduce cost, and is more conducive to industrial application process. The present invention also provides a method for using xanthine dehydrogenase to detect xanthine/hypoxanthine content in samples including serum and urine. The invention further provides a detection kit for detecting the content of xanthine/hypoxanthine in a sample. The detection method and the detection kit provided by the invention are not affected by the dissolved oxygen in the sample, have a wider detection range and higher sensitivity than the prior art, and are convenient for popularization and application. |
