abstract The invention provides benzothioxanthene derivatives and a preparation method and application thereof. The six benzothioxanthene derivatives containing different side chains provided by the present invention all have large conjugation planes and side chains that can be protonated containing three methylene groups, and have a certain effect on G-quadruplexes. Affinity and selectivity, can induce guanine-rich sequences to form a G-quadruplex structure, and the six benzothioxanthene derivatives can inhibit A549 cells and SGC-7901 cells to varying degrees granzyme activity; moreover, it can suppress tumor cell growth and promote its apoptosis by down-regulating the expression ability of PD-1 gene in JF305 cells and Bel-7402 cells, so it can be used to prepare genes for tumor therapy targeted drugs.
