abstract The invention discloses a preparation method of high-purity triglyceride type PUFA (Polyunsaturated Fatty Acid). The preparation method comprises the following steps: (1), using lipase Novozym435 as catalyst, enabling acyl donor of the PUFA to react with glycerol; removing catalyst and residual glycerol to obtain a mixture of trimester type PUFA and meta-glyceride PUFA; (2), carrying out hydrolysis or alcoholysis reaction to the meta-glyceride PUFA and acyl donor in catalytic mixture at a temperature not higher than 5 DEG C through the meta-glyceride lipase SMG1; (3), separating the reaction product to recycle oil phase, and purifying the oil phase to obtain the high-purity triglyceride type PUFA. The preparation method uses meta-glyceride lipase to selectively remove non-triglyceride components in the PUFA triglyceride at a low temperature to obtain a PUFA glyceride product with a low peroxide value and triglyceride content of 95-100%, and has better economical efficiency and environmental protection capability.
