abstract The invention discloses a cyclo-heterocyclic ketene aminal compound containing cyano-group and having a chiral center, and a synthetic method thereof. The structure of the cyclo-heterocyclic ketene aminal compound containing the cyano-group and having the chiral center is as formula I, wherein in the formula I, Z is NH, NR', O or S; n is 1, 2 or 3; R is aroyl, heterocyclic aroyl, fused ring aroyl, alkanoyl, ester group, nitryl or cyano-group; R' is aryl, heterocyclic aryl, fused ring aryl, alkyl, substituted alkyl, naphthenic group or substituted naphthenic group; and R' is alkyl, substituted alkyl, aryl or substituted aryl. The method uses simple and easily obtained heterocyclic ketene aminal or analog thereof and alpha-beta-unsaturated compounds with chiral assistants to carry out 1, 4 Michael addition, and carry out intramolecular amide aminolysis at the same time to form a cycle shape, so as to obtain a target compound (formula I) through further reaction. The compound can be used as an intermediate compound for synthesizing other heterocyclic compounds. The method has the advantages of simple synthesis technology, high yield and stable product. The method achieves parallel and efficient synthesis of the cyclo-heterocyclic ketene aminal compound containing the cyano-group and having the chiral center, and has the characteristics of high yield, simple courses, quickness and the like.
