abstract |
The present invention relates to acylated insulin analogs with extended duration of action. These insulin analogs or their pharmaceutically acceptable salts have the formula SEQ ID NO: 1 Xaa He Val Glu Gin Cys Cys Thr Ser He Cys Ser 1 5 10 Leu Tyr Gin Leu Glu Asn Tyr Cys Asn 15 20 and SEQ ID NO: 2 Xaa Val Asn Gin His Leu Cys Gly Ser His Leu Val 1 5 10 Glu Ala Leu Tyr Leu Val Cys Gly Glu Arg Gly Prh 15 20 Phe Tyr Thr Xaa Xaa Thr 25 30 and are properly cross-linked to each other, where Xaa at position 28 SEQ ID NO : 2 represents acylated Lys, and at position 29 of SEQ ID NO: 2, Xaa represents Pro. The acylating group is C6 - C17 fatty acid. The invention further provides a pharmaceutical composition comprising an effective amount of an acylated insulin analog of the present invention in combination with one or more pharmaceutically acceptable excipients and a process for preparing the composition. |
