titleOfInvention Method for identifying and representing interindividual differences of dna sequences
abstract The invention relates to a method for identifying and representing interindividual differences of DNA sequences. The invention is characterized in that; the genomic DNA of the individuals to be examined are isolated; segments of the genes to be compared, genomic regions, and genomes are defined; these segments are isolated in a targeted manner by means of PCR while using highly specific oligonucleotides and are produced in a sufficient number of copies; a simultaneous sequencing of all obtained gene segments is carried out, said gene segments now being available as PCR products; afterwards, the mixture of termination reactions which is produced thereby and which contains the individual sequences of all gene segments is electrophoretically separated on a sequencing gel; the mixture is transferred onto a membrane by means of direct transfer electrophoresis; afterwards, the individual sequences of a determined gene segment from the original mixture are made visible by hybridizing the membrane with a marked, highly specific oligonucleotide; this process is repeated unit all gene segments from the common pool are detected; afterwards, interindividual differences of DNA sequences are detected by image analysis, and a genetic profile model is established and evaluated by mathematical processing.
