abstract A method for determining the base sequence of a target DNA fragment which comprises amplifying the DNA fragment while forming a nucleic acid transcript by using an RNA polymerase with the use of the thus amplified DNA fragment as a template in the presence of a nucleic acid transcript terminator, and then analyzing the nucleic acid transcript thus obtained, wherein the amplification of the target DNA fragment and the formation of the transcript are effected isothermally. The amplification of the target DNA fragment and the formation of the transcript are effected at around ordinary temperature and the target DNA fragment can be amplified by the strand replacement/amplification method. According to this novel method, a DNA can be sequenced by effecting the amplification of the target DNA fragment and the formation of the nucleic acid transcript at the same time without resort to any thermostable RNA polymerase.
