abstract |
A method for preparing a retrovirus vector having a high titer in order to transfer desired foreign genes into target cells followed by the expression of the same. A DNA construction with the orientation of a promoter, an loxP sequence, a VSV-G gene and a polyA addition signal in this order is transferred into a cell carrying a retrovirus gag and pol gene expression system. Next, a retrovirus vector having the desired foreign gene is transferred thereinto followed by the treatment with a recombinase to give a pseudo-type retrovirus vector having a high titer. |
