abstract | The present invention provides an in vitro and in vivo bioassay utilizing mammalian tissues such that the action of agents that are effective in blunting the impact of hyperglycemia can be detected. The assay can be completed in approximately 10 days, is technically simple, has exquisite sensitivity and produces a graded response such that therapeutic agents can be identified and their relative efficacies can be assessed. Compounds exhibiting a positive result in this bioassay have the potential of being preventative agents in the following disease processes: diabetes (type I and type II), atherosclerosis, birth defects, and the recurrence of certain cancers. In employing the in vitro bioassay of the present invention, a class of compounds, folates, folate derivatives and pterins, has been identified that prevents the measurable biological consequences of hyperglycemia in a mammalian model. This novel class of compounds has not been linked previously to diabetes. Folates, folate derivatives and pterins have very low toxicity and make excellent preventative therapeutics. |
