abstract | Methods for purifying enzymes, in particular, acylases characterized by selectively aggregating and precipitating contaminating enzymes, in particular, deacetylases with the use of surfactants, in particular, cationic surfactants; methods for regenerating immobilized enzyme carriers, in particular, those made from synthetic absorbents or ion exchange resins by treating immobilized enzymes prepared by bonding the enzymes to the carriers optionally followed by cross-linking with proteases; methods for regenerating carriers as described above wherein the carriers are porous ones; and methods for regenerating carriers as described above wherein the enzyme is a cephalosporin C acylase. By the above methods for purifying enzymes, contaminating enzymes which are undesirable for the target enzymes but cannot be eliminated by the conventional separation techniques can be selectively eliminated. Thus, these methods are useful in the production of highly pure enzyme specimens. Compared with the conventional methods, the above-mentioned methods for regenerating immobilized enzyme carriers make it possible to very efficiently eliminate the enzymes from the carriers, in particular, porous ones through digestion with proteases. Therefore, the carriers can be used repeatedly, which makes these methods useful from the viewpoints of the environment and cost. |
