abstract | The application identifies a problem in the expression of transgenes in transgenic animals which renders the use of transgenic material unsuitable for successful xenograft transplantation. The difficulty identified is the varigated expression of the transgene which renders the xerograft vulnerable to hyperacute rejection after implantation. The solution described is to include a region corresponding to a substantial portion of the region naturally upstream of the gene of interest. Preferably the region includes the locus control region (LCR) for the gene of interest, preferably together with the intervening section which naturally links the LCR to the gene of interest. The transgene construct described is therefore extremely large and is desirably manipulated and amplified using YAC technology. |
