abstract A process is disclosed for isolating and purifying nucleic acids and/or oligonucleotides for gene therapy. The nucleic acids and/or oligonucleotides are isolated or purified from a substantially biological source. The process is characterised in that the substantially biological sources are disintegrated, if required the residues of biological source are removed or eliminated from the thus obtained fractions by a mechanical process known per se, such as centrifugation or filtering, the thus processed fractions are treated with affinity chromatography material or with inorganic chromatography material for removing endotoxins, the nucleic acids and/or oligonucleotides are isolated on an anion exchanger designed so that DNA starts to be desorbed from the anion exchanger only when the sodium chloride solution ionic strength is at least about 100 mM higher than the ionic strength at which the RNA of the anion exchange material starts to be desorbed from the anion exchanger.
