titleOfInvention | Reference control for use with manual and flow cytometric reticulocyte counting devices |
abstract | A stable reticulocyte reference control which is usable with both manual and flow cytometric reticulocyte counting devices and methods such as those used to access bone marrow activity is provided. The reference control includes a suspension of erythrocytes that are loaded with either nucleic acid or a polyanion capable of binding with a cationic dye, in an aqueous preserving medium. The reference control is preferably prepared using a reversible osmotic lysis procedure whereby the nucleic acid or polyanion is encapsulated inside the erythrocytes by first introducing the cells into a hypotonic solution in the presence of the compound to be encapsulated, followed by replacing the hypotonic solution with a hypertonic solution. The reference control of the invention is advantageous in that it has long-term stability and it can be used to give highly accurate readings of reticulocyte counts using either manual or flow cytometric procedures. |
