abstract The present invention relates to poly- and oligonucleotides, particularly to DNA and RNA probes and PCR primers, methods of their production and their use in restriction fragment length polymorphism (RPLP) and minisatellite variation repeat mapping analysis (MVRM). The probes of the invention have particular application as single locus probes for use in genetic characterisation techniques. The probes and primers have a nucleotide base sequences capable of specific hybridization with a sequence comprising a sequence (I): 5' (CCCCT CACTG TCCAC (L)CTC(M) TG(N)CC TATAG AGAC)n 3' or its complementary sequence (II): 3' (GGGGA GTGAC AGGTG (M)GAG(L) AC(Q)GG ATATC TCTG)n 5' or sequences having up to 15 % mismatch to them; wherein L represents T or C; M represents A or G; N represents G or T; and Q represents C or A; n is an integer from 1 to 1000 and 5' and 3' refer to the respective 5' and 3' ends of the sequence. T is thymine or uracil and bases may be replaced by those having equivalent base pairing characteristics.
