titleOfInvention | Submicron particles, preparation and utilization in immunodiagnosis |
abstract | The present invention relates to a method for fixing by covalence or adsorption an immunologic material selected amongst antigens and antibodies on submicron particles of latex, said method, which includes the stabilization of the latex, being characterized in that it is comprised of (A) contacting in an appropriate liquid medium having a ionic force in the range from 0.01 to 0.5 and a pH from 4 to 10, at a temperature from 0 to 60°C (1) particles of acrylic latex based on butyle polymethacrylate, having a mean diameter lower than or equal to 100 nm, said latex containing a molar quantity of butyle methacrylate patterns higher than 50 %, with a density from 0.9 to 1.4 g/cm3 and including from 100 to 400 and 450 microequivalents of groups COOH per gram of latex, with (2) the immunologic material in order to obtain latex particles sensitized by an immunologic material and such that said immunologic material is bound to each latex particle, and (B) stabilizing said immunologic reagent thus obtained by means of a stabilizing material selected amongst the group including hydroxylated substances containing one or better a plurality of groups OH per molecule, proteinic substances, peptidic substances, aminoacids, polyozoïc substances including one or a plurality of groups carboxylic acid and/or carboxylate, polyvinylpyrrolidone and mixtures thereof. The invention also relates to the immunologic reagent obtained according to such method and its utilization in the field of immunologic dosages. |
