titleOfInvention Cell-seeding into fibrous lattices by means of centrifugation
abstract The present invention includes the use of centrifugal force to introduce viable cells into a fibrous mesh, as well as fibrous mesh which are seeded with cells by the use of centrifugal force. A variety of fibrous mesh can be seeded by the methods of the present invention, such as highly porous mesh comprising collagen fibers crosslinked with glicosaminoglican. Prior to centrifugation, a piece of intact tissue is collected from a donor site. It is treated with one or more substances, such as trypsin or collagenase, to dissociate cells from tissue. The cells are then mixed with an aqueous solution to create an aqueous suspension of cells. A piece of the fibrous mesh is placed inside a container, referred to herein as a "bucket", indicated to be driven in rotation by a centrifuge. The aqueous suspension of cells is placed inside the well, in contact with the mesh. The centrifuge is then activated. The centrifugal force causes a relatively uniform forced distribution in the mesh of cells which are denser than the aqueous solution. By controlling various parameters the cells can be seeded at any desired location of the trellis.
