titleOfInvention Monocloal antibodies specific for human hematopoietic cell surface glycoproteins
abstract Monoclonal antibodies are produced for a family of glycoproteins which are selectively expressed on the surface of human nucleated hemopoietic cells but which are absent in other normal human cells. Mice are inoculated with human hemopoietic cells or fragments thereof, and spleen cells obtained from mice are fused with mouse myeloma cells to produce hybridomas. Hybridomas are grown as clones, and antibodies obtained from individual clones are tested to determine their specificity for hemopoietic cells. Clones which produce antibodies specific for the surface glycoproteins of human nucleated hemopoietic cells are selected for subsequent culture to produce the antibody, and the antibody is obtained from a culture growth medium or from the fluid ascites in mice containing the hybridoma tumor.
