abstract | Improved process for the isolation of amino acylase from the kidneys of mammals. According to the invention an aqueous extract of kidneys, prepared according to a known process, is subjected to a heat treatment at a temperature ranging from 60 to 80 C for 5 to 15 minutes, the mixture treated by heat is centrifugal, 200 to 300 g / liters of a salt corresponding to the general formula KxA (in which K represents a monovalent cation, A represents the anion of a mineral acid and x is equal to the valence of the anion) are added to the supernatant, the suspension obtained is centrifuged, the separate precipitate is dissolved in water, the solution is dialyzed against water and, optionally, the dialyzed solution or a solution of the enzyme, separated from the dialysate, in a buffer with a pH ranging from 6.5 to 8.5 is subjected to anion exchange chromatography, and finally, optionally, the active fractions obtained in the chromatographic step or a solution of the enzyme, precipitated from the active fractions by means of a salt corresponding to the general formula KxA, in a buffer of one pH values from 6.0 to 8.0 are subjected to gel chromatography on a filter in which the upper limit of the molecular weight of fractionation is higher than 70000, and the enzyme is precipitated from the active fractions by means of a salt corresponding to the general formula KxA already described.
