Plasmid, stem cell and application |
abstract |
Provided are a TALEN/TALENickase-mediated non-viral gene targeting plasmid and an iPSCs stem cell having an N-terminal for fusion of an isoleucine zipper and integrated with a TRAIL expression box and an application of the stem cell. The sequence of the plasmid is shown as SEQ ID NO:1, and the plasmid is combined with a gene editing tool TALEN to achieve site-specific integration of a therapeutic gene TRAIL. TRAIL-iMSCs sustainably express TRAIL, and have the effect of inducing apoptosis on multiple types of tumor cells. |
