abstract Described herein are the discovery and/or optimization of enzymes involved in the biosynthesis of olivetolic acid, divarinic acid, and analogs thereof. Described herein is the discovery and/or optimization of enzymes involved in the biosynthesis of Olivetolic acid (OA) from hexanoic acid. Specifically, disclosed are engineered enzymes, cells, and methods that significantly enhance both the rate and total quantity at which cannabinoid precursors can be biosynthesized. This is accomplished by increasing the flux of required precursors for CEGAS activity, olivetolic acid (OA) and geranyl pyrophosphate (GPP).
