abstract | This method for producing a synthesized DNA molecule constituted of a desired sequence includes: a step for preparing a plurality of template oligonucleotides each having a portion of the desired sequence; a step for preparing a plurality of splint oligonucleotides each having, when the plurality of template oligonucleotides are arranged in a desired sequence, a sequence complimentary to the sequence of the end portions of the adjacent oligonucleotides; a step for annealing the plurality of splint oligonucleotides to the plurality of template oligonucleotides; a step, which is carried out after the step for annealing, for ligating the plurality of template oligonucleotides to one another, and synthesizing a circular single-strand DNA that includes the plurality of splint oligonucleotides as complementary strands; and, with the circular single-strand DNA as the template, a step for synthesizing a double-strand DNA using φ29 DNA polymerase. |
