abstract | Exemplary embodiments of the present disclosure are directed towards primers for differential detection of fecal coliforms in a water sample by multiplex PCR and uses thereof. The method comprising: 194F, 474F and 1436R three primers designed for specific amplification of V2, V3,V9 hyper variable regions of 16S rRNA gene. Multiplex PCR allowed for differentiation of the total coliform into fecal coliform and non-fecal coliforms by specific amplicons size: 1285 bp amplicon is specific for 6 non-fecal coliform genera and 1009 bp amplicon is specific for fecal coliform i.e., E. coli. If the drinking water was contaminated with both fecal and non-fecal coliforms, two amplicons of 1285 bp and 1009 bp was observed. The kit enabled rapid identification of the total coliforms in the large number of water samples compared with traditional methods. Results can be acquired within 3 hrs of time as compared to 3-4 days in MPN method. |
