abstract Provided are methods for generating a sequencing library from a sample comprising a plurality of mammalian cells. Also provided are methods for measuring protein-chromatin interactions in mammalian cells. The disclosed methods comprise various steps such as incubating with beads a plurality of nuclei, fixing, restriction digesting, proximity ligating, generating DNA-antibody mixture, performing a nucleic acid amplification to generate a sequencing library and optionally producing a plurality of sequencing reads.
