abstract The present disclosure relates to a method of isolating and processing antibody-specific B cells from a subject, comprising: providing antibodies from a blood sample from said subject covalently or non-covalently linked to a surface; contacting said antibodies covalently or non-covalently linked to said surface with a peptide display library under conditions sufficient to permit binding of an antibody linked to said surface to an antigen within said peptide display library, thereby isolating said antigen; contacting said antigen to a sample comprising peripheral blood mononuclear cells (PBMCs) from said subject under conditions sufficient to permit binding of an antibodyexpressing PBMC from said PBMCs from said subject to said antigen, thereby isolating said antibody-expressing PBMC; and amplifying at least one immunoglobulin sequence expressed by said isolated antibody-expressing PBMC. In addition, the invention is directed to an isolated PBMC isolated by the method.
