abstract |
Provided are a porcine alveolar macrophage healthy cell line, and a construction method and an application. The porcine alveolar macrophage healthy cell line has an accession number of CGMCC No: 21498. In the porcine alveolar macrophage healthy cell line, a 100% healthy porcine alveolar macrophage cell line is screened by using a special cell culture solution in combination with a natural domestication selection method; the porcine alveolar macrophage healthy cell line can be applied to cell culture of viruses, research and development and production of live vaccines and inactivated vaccines and preparation of bioactive substances secreted by M1 and M2 macrophages after induced polarization to be respectively used for cancer treatment, inflammation diminishing and the like. The construction method provided does not employ any carcinogenic substances such as lentivirus transformation or plasmid transfection, and the constructed porcine alveolar macrophage healthy cell line keeps the characteristics of primary porcine alveolar macrophages. For example, in terms of sensitivity to porcine virus infection, the increasingly difficult research of using a Marc145 cell line to separate and culture a newly emerging porcine reproductive and respiratory syndrome virus, and using a Vero cell line to separate and culture a newly emerging virus such as a newly emerging porcine epidemic diarrhea virus mutant strain becomes easier. |
