abstract | The invention relates to an in vitro method of detecting by RT-qPCR on a sample, presence of a nucleic acid molecule contained in the mitochondrial Small Sub-Unit (mtSSU) gene of Histoplasma capsulatum, or a variant thereof, by Whole Nucleic Acid (WNA) reverse transcriptase qPCR (RT-qPCR). The method can be coupled to an in vitro method of detecting nucleic acid content of Mycobacterium tuberculosis in particular by RT-PCR. The invention also relates to an in vitro method of diagnosing whether an animal or human subject is infected with Histoplasma capsulatum, or has histoplasmosis, optionally whether the animal or human subject has or has a risk to develop tuberculosis, based on the detection method of the invention. The invention can be applied to determining the efficacy of a therapeutic active ingredient. Are also encompassed therapeutic applications for patients diagnosed as having histoplasmosis with the method of the invention, kits and their uses for carrying out a method of the invention, primer nucleic acid molecules. |
