abstract A novel chimpanzee adenovirus vector, a construction method therefor, and an application thereof. The chimpanzee adenovirus vector has higher virus titer, and a measurement method for the virus titer thereof is provided. An annular chimpanzee adenovirus vector is constructed by means of a shuttle plasmid, and E1 and E3 are knocked out by means of CRISPR/Cas9, thus a replication-deficient chimpanzee adenovirus vector is constructed. The chimpanzee adenovirus vector has no pre-existing antibody in people; E1 and E3 knockout is secure, and is significantly different from human adenovirus 5 type E1 in 293 cells; the chimpanzee adenovirus vector can greatly avoid recovery mutation (RCA) and is more secure. A constructed novel coronavirus vaccine is strong in vaccine stability after dozens of generations, does not cause mutation, and can induce strong humoral immunity and cellular response in a mouse model.
