abstract Provided are a carrier comprising a temperature switch loop and application thereof in increasing the yield of threonine in microbial cells. According to the carrier, heterologous expression of pyruvate carboxylase is controlled, temperature-sensitive and easy-to-decarboxylate chemical properties of oxaloacetic acid are combined, metabolic flux is rebalanced between pyruvic acid and oxaloacetic acid, a central metabolic pathway is dynamically regulated, and supply of reduced cofactors is guaranteed, so as to promote production of L-threonine.
