abstract A method of producing robust and pure mesenchymal-like stem cells derived from induced pluripotent stem cells is disclosed. A population of induced pluripotent stem cells (iPSC) is differentiated in suspension without a feeder layer or imatrix. The differentiated cells are passaged in the presence of a mesenchymal stem cell (MSC) medium for a time and under conditions sufficient to produce mesenchymal stem cells. The differentiated cells are maintained in the culture medium for long term with no sign of epigenetic memory, thereby producing robust and pure mesenchymal-like stem cells. The method utilizes iPSCs as the starting material, due to their ability to proliferate indefinitely and create an essentially infinite amount that could be differentiated into extremely large quantities of MSCs. This enables to form a single master cell bank of iPSCs derived from a single donor to manufacture all of an ever-needed population of MSCs.
