Provided are a D-psicose 3-epimerase mutant, and an application thereof for preparing D-psicose in D-fructose isomerization under microbial catalysis. The D-psicose 3-epimerase mutant is obtained by subjecting the amino acids as represented by SEQ ID NO: 5 to site-specific mutagenesis of amino acids at one or more of the 242nd site, the 105th site, the 210th site, the 147th site and the 184th site. The mutant has a high optimal reaction temperature of 85°C, a highest product yield can reach 40.1%, which is better than the conversion effect of original enzymes and other mutant enzymes, and the mutant has a good industrial application prospect. |
