abstract Peptide antagonists of MDM2 and MDMX are disclosed. The peptide antagonists include antagonists of MDM2 or MDMX, as well as dual-specificity antagonists where the same peptide is an antagonist of both MDM2 and MDMX. The peptide antagonists have at least nanomolar binding affinity for MDM2 and/or MDMX, and in some cases picomolar binding affinity. The peptide antagonists can be used in methods based on blocking inhibition of p53 activity by MDM2 and/or MDMX. Such methods can be used in the treatment of diseases, such as cancer.
