The present invention relates to a method for detecting off-target sites of one or more target-specific programmable nucleases in a genome in vitro, which allows for the detection of off-target sites in an input sample of less than 1 µg of genomic DNA. The present invention also relates to a method for detecting in vitro off-target sites by using the in vitro method for detecting off-target sites of one or more target-specific programmable nucleases in a genome. |
