abstract | The present invention provides a preparation method for an RNA library, a sequencing method and a kit. Disclosed in the present invention is a method for preparing an RNA library. The method comprises: generating first strand cDNA by means of inverse transcription by using RNA as a template, to obtain an RNA/cDNA hybrid; and adding a first preset sequence to at least one end of the RNA/cDNA hybrid to form an RNA/DNA hybrid, the RNA/cDNA hybrid being an RNA library, and the first preset sequence being located on at least one end of a cDNA strand. This method is simple and quick to prepare an RNA library, and the constructed RNA library can be directly used for sequencing. Because this method has no second-strand cDNA synthesis process, that is, no cDNA amplification and enrichment process, bias and errors caused by PCR amplification and enrichment can be avoided. In addition, disclosed in the present invention is an RNA sequencing method, for directly determining cDNA sequence information to obtain RNA sequence information. Also disclosed in the present invention is a kit for RNA sequencing. This kit can be utilized to construct an RNA library and used for the sequencing of the RNA library. |
