abstract The present invention provides, in order to obtain a uniform quality for heavy-chain antibodies, a heavy-chain antibody that is less susceptible to modification by O-linked sugar chains. The heavy-chain antibody according to the present invention is characterized by including a heavy chain variable domain having a structure of (FR1)-(VH-CDR1)-(FR2)-(VH-CDR2)-(FR3)-(VH-CDR3)-(FR4), wherein, in the amino acid sequence of FR3, at least one serine or threonine residue is substituted with an amino acid other than serine or threonine. The present invention also pertains to: a nucleic acid having a base sequence encoding the amino acid sequence of the heavy-chain antibody; and a vector and a host cell including said nucleic acid.
