abstract | A method for improving quality and transplanting survival rate of Atractylodes lancea test-tube seedlings, comprising the steps of taking apical buds of robust Atractylodes lancea plants as explants, after cleaning and preliminary disinfection, immersing them in an antibacterial agent, taking them out and rinsing, then inoculating them in a buffer culture medium to obtain aseptic seedlings; transferring the aseptic seedlings into a proliferation culture medium for cultivation, transferring once every 28-30 days, continuously transferring for 4-5 times, so as to obtain cluster buds of Atractylodes lancea; transferring the cluster buds of Atractylodes lancea into a strong seedling rooting culture medium for cultivation, selecting the Atractylodes lancea test-tube seedlings with a height of 4-6 cm and 5-6 roots, moving them into outdoor for open-lid domestication culture, washing away the residual culture medium, immersing them in a carbendazim solution, and finally transplanting them on a seedling tray. The method takes the apical buds as the explants to establish an aseptic line, simplifies the steps of the Atractylodes lancea tissue culture and rapid propagation system, shortens the seedling cycle of test-tube seedlings, saves costs, and the obtained Atractylodes lancea seedlings have a high transplanting survival rate, and stable and excellent quality. |
