Disclosed are a heterodimeric fusion protein, a preparation therefor, and an application thereof. A heavy chain and a light chain of a first antigen binding domain Fab are respectively fused to an N terminal end of two Fcs, and on this basis, a second antigen binding domain, scfv, Fab2, Fv, a nano-sized antibody or a physiologically active peptide is fused to an N terminal end of the Fab or a C terminal end of the Fcs. The heterodimeric fusion protein prepared by the above method has the following benefits: each antigen binding domain in the molecules thereof has the same structure as the protein itself, such that the activity and the pharmaceutical properties of the antigen binding domain can be well retained; at least one antigen binding domain can bind to an antigen monovalently; and since an Fc fragment is retained, the heterodimeric fusion protein of the present invention has a prolonged half-life. |
