abstract | A gene synthesis method. Said method comprises: segmenting multiple target genes into several overlapping oligonucleotide sequences, adding a restriction enzyme cutting site sequence and a tag sequence at the end of the overlapping oligonucleotide sequences, the tag sequence being different due to different target genes; specifically linking the overlapping oligonucleotide sequences in the form of a single-strand and linked with the tag sequence to a magnetic bead of a reverse complement linked with the tag sequence by means of a double-stranded structure, then performing restriction enzyme cutting in a water-in-oil system to obtain single-stranded overlapping oligonucleotides, and then performing polymerase chain assembly, so as to obtain multiple target gene fragments at the same time. |
