abstract The present disclosure relates to the construction of a Mucor circinelloides cell plant for producing stearidonic acid SDA and fermentation technology thereof, and belongs to the field of genetic engineering. The present disclosure obtains recombinant strain Mc-Δ15 by cloning Δ15Des gene from Mortierella alpina, connecting same to an integrated plasmid pMAT1552, transforming said plasmid into Mucor circinelloides deficient strain Mu402, and integrating Δ15Des into the genome of Mucor circinelloides by means of homologous recombination, and finally achieves expression of the Δ15Des gene in Mucor circinelloides. There is no significant difference in the grease content of the recombinant strain Mc-Δ15 compared to control strain Mc1552. However, the grease composition changes greatly, and SDA is present in the grease of expression strain Mc-Δ15, and the content thereof reaches 5.2% of the total fatty acid content. By optimizing fermentation conditions and adding precursor fatty acids, the SDA content reaches 8.3%. The new recombinant strain was deposited on 20 June 2018 in China General Microbiological Culture Collection Center located at No. 3, Courtyard No. 1, Beichen West Road, Chaoyang District, Beijing. The deposit center gives the biological material with the deposit number CGMCC No. 15888, and the suggested classification name is Mucor circinelloides-D15D.
