abstract | Provided is an application of two genes sirtuin2 and sirtuin4 and proteins thereof in Rhodotorula glutinis for improving mitochondrial function, pertaining to the technical field of biology. Recombinant expression, in mammalian cells, of the genes sirtuin2 and sirtuin4 in Rhodotorula glutinis having functions of reconstructing the mitochondrial network and improving the phenotype of mammalian senescent cells is provided. Experimental results showed that the genes can activate damaged mitochondrial autophagy-related proteins, significantly reduce the activity levels of SA-β-gal in mammalian cells, enhance the expression levels of CK19 and β1 integrin, reduce the levels of intracellular ROS, significantly increase ATP levels in the cells, and improve mitochondrial morphology, thereby improving the mitochondrial network and increasing the ability of the cells to resist aging. An application of the genes sirtuin2 and sirtuin4 in Rhodotorula glutinis and proteins thereof in the preparation of a medicament for preventing and/or treating mitochondrial dysfunction or a hypofunction disease or pathological process is provided. |
