titleOfInvention Method for encapsulating active substances agents in liposomes
abstract The invention relates to a method for encapsculating active ingredients agents in liposomes, which include an active agent solution encapsulated by with a bilayer formed by two monomolecular layers formed by amphiphilic compounds, wherein the method comprises the following steps: (a) providing the active agent solution; (b) providing an emulsion by emulsifying the active agent solution in a first fluid liquid that cannot be mixed with the solvent thereof in the presence of the amphiphilic compound, obtaining pre-liposomes; (c) providing a liquid phase that cannot be mixed with the first liquid of the emulsion; (d) bringing the emulsion in contact with the liquid phase, forming a phase boundary, in order to enrich the amphiphilic compound here; and (e) centrifuging the contacting emulsion and the liquid phase, which are in contact with one another via the phase boundary, in order to transfer the pre-liposomes out of the emulsion into the liquid phase, wherein, whereinwhen passing the phase boundary, the amphiphilic compound enriched there accumulates on the monomolecular inner layer of the pre-liposomes, forming a monomolecular outer layer, in order to generate the bilayer. In order to prevent the formation of organogels as a result of an excessive accumulation of the amphiphilic compound at the phase boundary, according to the invention, the first liquid of the emulsion is selected in such a way that the solubility of the amphiphilic compound in the first liquid is max. 1 x 10-4 mol/l.
